End of Gathering Storm III when Gulliman, Cypher and Co. finally reach the Imperial Palace. Gulliman winds up backing out of a deal he made with Cypher where he'd allow him an audience with the Emperor (as he recognised his sword and thought letting him in the same room as Emps PROBABLY isn't a good idea) and subsequently ordered the Custodes to restrain and place the rogue astartes in a guarded cell that has not been breached since its creation over ten millenium ago.
Cypher predictably is shown gone in the next page over.

The problem with choosing Cypher is he counts as one of the few characters with a proven 0% success rate in getting into the emperor's presence.

That's a bit ridiculous though. Chaos can't persuade you to do anything. They can only offer you something and if you take what they offer your soul is taken. Chaos can't dupe you into turning, espeacially not the Custodes who know as much as the Inquisition does about Chaos.
Chaos is all about lies and trickery. Schemes and schemes within schemes. Some more than others. And anything with a soul can be tempted in some way. The custodes have all these titles and trials which suggests that there is an ego there of some sort. Chaos can always find a way. Ask Horus and the word bearers about chaos lying. Or even the emperor if the tales are true.

I think Guilliman would have the best chance since I think he's likely the only one who has managed to be alone with the Emperor, not that he would though.

A Custodes being incorruptable doesn't mean one couldn't be used to assasinate the Emprah.
In theory, some of the craftier ones (Tzeench, Cegorath, probably nothing short of that level) could connive such that a guard or tech learns what they're supposed to do ever-so-slightly differently than they should have, and maybe even a couple of them performing their duties in the wrong way in just the right alignment and... oops, no more Emprah.
As for actually getting into the throneroom short of sacking Earth itself, the only possibilities I see are:
-Vect himself, using much of his political capitol to arrange for things and get stuff like phase tech
-Harlequins under Cegoraths orders, with a great deal of effort and some luck
-Particularly advanced and desperate Necrons with phasing tech and minor miracles
-Cypher, because plot armor. He's faster than Bahroth!
