For example, a unit of space marines is within range of 2 Icons of despair, which give a -1 to leadership rolls. Would this unit suffer -2 to it's leadership rolls?

It depends on the wording of the specific rule. For example, two Icons of Despair do not stack due to their wording.

Like Ghaz said, there is no one size fits all answer. It all depends on the wording of the rule, and whether it says it applies only once or not.

Generally, ones that do not stack will say "If X is in range of any Y" rather than "a Y".

Yep, the rules for stuff like this will general read something like:

"If a unit is within 6" of an Icon they will suffer -1 to their leadership..."

OR

"For each icon within 6" of a unit, that unit suffers -1 leadership."

I'm paraphrasing here, but you get the point. It will explain (normally) if it occurs multiple times or not.
