I believe there are far better writers on this board than there are for the splatter movies going these days. With this in mind, I'm starting this thread so we can get an ongoing splatter plot going that will put anything out there to shame.

Good idea, Relapse. I've always wanted to write some supernatural horror, in the manner of Stephen King or Elm Street. I should have something to show tonight (that'll be a while however, as it's only 1:05 PM here)

When I was in high school I, loosely, reinterpreted Hamlet as a slasher flick. It wasn't very good, because I was like 16 when I wrote it, but the idea has stuck with me.
Othello would also be a good choice. As would Titus Adronicus (obviously).

I can see that working quite well, but I would probably work it further, into a splatter-fest flick, skipping the rather unnecessary nuances that a slasher would provide; using the script to convey that sublayer more effectively.
Lots of spray and red stuff, no discreet references, just rather direct interpretation, combined with some witty story telling; and the blood does the rest of the talking. Shadows, highlights, all that stuff, would deal rather well with the more obtuse emotions conveyed by the master works.

Well, I pushed the setting forward to Al Capone's Chicago. The titular character wielded a meat hook in the opening scene, as opposed to a halberd. The whole thing smacked of the DiCaprio Romeo & Juliet (can't remember the name of the director), but with a decidedly bloodier angle. I did some rather interesting things, like allude to the madness being the result of syphilis, but otherwise it was largely faithful. The 'loose' component was primarily one of script.
