So I've entered my list so I can't modify it so you can't help with that, however I'm unfamiliar versions Tyranids, Dark Eldar, Sisters and against other Eldar. I'm not sure or their lists but what am I likely to face and how should I counter it? Here's my list

Autarch, Jetbike, laser lance, fusion gun, banshee mask, Mantle of the laughing God
10 Harlequins, 8 kisses, 2 fusion pistols, shadowseer, troupe master, death jester
10 Guardians, wave serpent w scatter laser and shurikan cannon
5 dire avengers, wave serpent w scatter laser and Shurikan cannon

So are the armies you mentioned in the tournament? Or do you know the other armies?

Because none of those should cause you much trouble.
Tyranids - sit back and fire. Take of the Synapse Creatures (aka the big ones) with spammed S6.
After that they are hardly a problem.
Dark Eldar - tanks and lots of them. Your Autarch might prove handy in popping them, just like your Fire Prism.
Might be challenging if played well.
Don't know much about Sisters of Battle but I would expect Rhino-based tanks. Your firepower should work against them.

But if there are other Eldar armies you might be in problem.
Harlequins aren't generally that good and they rely on Shadowseer's psyking to survive.
Now if the enemy has Farseers, Warlocks etc he might easily deny that from you, making your pricey unit an easy snack.

With Tyranids, it's also worth noting that most of their weapons are very mid-ranged (12-18") but get extremely nasty once they are in range. So, you generally want to spend as little time inside that kill zone as possible. Harlequins will have to close in order to do damage, so they will be playing the Nids game. The WS and Guardians can keep their distance better (Guardians by just hanging back, if need be, DAs by abusing battle focus). Nids are not really the close combat team they're cracked up to be, so don't be afraid of charging them if you get the Harlies in range. The biggest threat to this list is going to be a swooping flyrant, as you don't have a lot of good answers to it. Weight of fire from the DAs, SS and Scatter Laser could do the trick. The other FMCs aren't that threatening to your list (although Harpy could be a problem, it is not that popular). Also, watch out for Biovores dropping pie plates on your troops, as they can put a lot of hits on unprotected infantry. If you see biovores, you may want to prioritize them as they are rather squishy and you should be able to reach them fairly easily with the Autarch/WS early on.

Targeting synapse is always good if you have nothing better to do, but keep in mind that it is not always as easy as Step One, kill synapse, Step Two, profit. I have won games where I had no Synapse creatures left because, surprise, Nids sometimes pass leadership checks and not all of the results on the IB table are bad (and sometimes they even get a bonus trait, such as Rage on the Feed table, IIRC). Yeah, Gaunts aren't going to stand up if they fail IB, but for something like a T-Fex or Carnifex, they can still do damage even after failing IB (assuming the Carnifex isn't in a brood and therefore getting into a headbutting match with its buddies).

I know there is no sisters of battle players, he's got stuff to do(excuse I bet), and 1 eldar who runs wave serpent spam, I was actually intending to bring my spiders instead of my harlies but because the spiders aren't painted very well and since a large portion of points is painting, my harlies will help in that regard.
