What is the most powerful melee weapon we have seen any fluff etc?
To get the stupid arguments about the wielder out of the way, let's act on the basis that all weapons are being used by the best possible master of said weapon.
When it kills something, it also destroys their warp presence. That means daemons are completely dead and there's no chance of chaos resurrections either for mortal men either.
Have to go with Lucius' lash as it causes the victim intense pain and if hes killed his soul passes on to the guy who ckilled him. He is literally invincible.

the immortality comes from Lucius though, not his whip.

Seriously? Where's that fluff from, because then yeah, I'd say the Lion Sword is pretty awesome. Useless against Necrons, and pointless against Tyranids though.
Crowe's blade of Antwyr which sounds pretty badass. If he was able to tap into its daemonic side he would probably be invincible but that would go against the purity of the GK.
It does pretty much what you would expect a blazing eldritch artifact said to contain the baleful energies of a dying sun would do- cut through a hive fleet like a Zamboni through a field of ice cream sandwiches

I'm going to have to go with that sword that wounds horus. I forget what it's called...but it is essentially a sword that produces a toxin specifically designed to kill its victim outright.

I killed lucius with a Wraithlord. What happens then?
I'd go with the spear of Twilight, or the Lion's sword.
a trusty axe or chain axe because...hey its simple it has none of this complex mumbo jumbo such as power weapons or phase weapons.
it just works & it does its job of hacking enemies to pieces
The problem with the Anathame is you need to know your opponents name for it to be a guided killing machine. This makes it good for the HH story since it's all brothers fighting amongst themselves, and even makes it good for assassin work.
As a general weapon, it becomes just another sword, since you're not going to know the names of every opponent on the battlefield, or even the big tough guys that you would want to single out with the blade.

Easy enough mistake to make, considering every single Khorne hero and character made by GW uses a bloody axe, and there used to be old wargear called Axe of Khorne.
True followers of Khorne should be emulating their god, and using a sword.

The Spear of Twilight. An spear that houses the powers of an dying sun, consumes the soul of its wielder to function and was used by an freaking eldar to kill an hive tyrant and its retinue! On the other hand, the gauntlets of ultramar or crowes own demon sword.
The Axe Morkai- because most of the bette ones have been taken already (this may have been posted before, but I cant be assed into reading 4 pages...).
Its a daemon weapon... stolen, then reforged into a GIANT FRICKING AXE, used by arguably one of the best leaders of the imperium.
Oh, I almost forgot about some of my personal favorites-
