Recently i participated in a 32-player tournament... this tournament was my first ever so i didnt have any high hopes of winning and sort of stuff.
So i made a fluffy theme list instead with an Exalted Hero on a Jugger as a general...

Anyhow since my whole army was devoted to khorne i got baited constantly and in my first game against Brets my Knights charged a unit of KotR who fled and got charged by another unit of KotR and a unit of Grail knights with a paladin and a bsb, and according to the Eye of the Gods i have to issue a challenge. So i did and he accepted with a regular Grail knight since all of them are champions. My knights actually pulled through and held and my Hero butchered the grail knight.

Now to the question... do i have to issue another challenge (in the same fight)? We went with that anyway and my hero stood there for the remainder of the game chopping grailknights and getting an overkill of 3-4 everytime. (my knights took down his paladin and bsb).

You have to issue a challenge at any point, and as long as a combat is continuing then there is space for a challenge - regardless of how many rounds the fight lasts for.

You may only have 1 challenge at a time, but nothing states you can only have one per fight.

you should read the FAQ too, all characters and champions in the chaos army must issue challenges if the opportunity presents itself (you can choose which one if there is more than one), but only those with the Eye of The Gods rule are prevented from refusing. Note that if you have a sorcerer and a champ and someone issues a challenge you can accept with the champ, so long as no one refuses while the EoTG character is alive and in the unit.
