Most powerful/accomplished/influential? That's a tricky question. Saint Dominica is said to be universally known across the Imperium and basically ended the reign of Vandire with a single sweep of her blade, in addition to being the founding Saint of the Sisterhood.

On the other hand, her Sororitas aren't really a vital force in the IoM, although they do fulfill an important symbolic function and may, at times, accomplish noteworthy feats, in addition to providing useful services to other Imperial bodies. One could probably say they are one of many pillars holding everything together, and like the Space Marines they are not irreplacable but very much "nice to have".
