The Emperor is an extremely powerful psyker, the most powerful human psyker to ever live. Easily assassinated by a Culexus? Probably not, because the Emperor probably foretells the assassin's arrival a week ahead of the event, and so the poor bastard dies in a freak speeder accident on his way to the job (or is accosted and killed by Custodes upon arrival).
That's the point. Culexus assassins are the ultimate counter to psykers. They are completely undetectable by psychic powers, meaning that future telling can't take them into account. Even against an Eldar they are nightmarishly potent assassins. It's in their very nature to grow in their own power while they are in presence of a psyker. Any Culexus assassin would simply be supercharged by sheer amount of energy which Emperor carelessly emits. (Remember novel ''Thousands sons'' and how Ahriman described the Emperor before trial. He was emitting his power like a supernova. Probably for showcase purposes, because a serious psyker would prefer hiding his presence). Even more, Black pariahs might just horribly kill Emperor by their mere presence alone. Even though, I fail to see how soulless pariah can be even more soulless...
Btw: I think that more authors should write stories as him(I have more in mind movies and games). I like unpredictable stories where you are hard-pressed to name good guys.
