So, I've begun collecting bikerboys for a Wazdakka list, but a situation came up in yesterday's game with my old shooty-mek list that made me think twice. My Dark Eldar opponent had a large squad of warriors in a four-storey building that was also his command base. Now I had a hard enough time dealing with all that gun fire against my boys, and the game ended in a draw before the actual assault occurred.
Question: if bikerboys were to want to assault the building but no troops were on the bottom floor, how or could my bikers reach the rapists inside? Can bikers go up a building?

Bikers cannot enter an intact building or go onto the upper levels of a ruin.
Since the first model to move into combat in the assault phase must move into base contact you would not have been able to assault.
This is a major disadvantage to an all-bike army.

You can always use the Nob withb the claw to attack the building itself. My bro plays orks and keeps on saying he is going to do this to me with my eldar pathfinders on a third floor. Have not presented the opportunity to him yet.

To clarify here... if it was a Building you couldn't have assaulted them directly anyway. You would have to attack the building in the hope of forcing them out. You also don't track what floor units are on... they're just 'inside' as they would be in a transport vehicle.
If it's a Ruin, then you could assault them if you could physically get into contact. The Ruins rules allow a certain amount of leeway for model placement, but you're still going to be hampered by the Bikers' inability to move above ground level.

Interesting idea.... I suppose they could hid from shot in the rubble?

As a further clarification... you can attack occupied Buildings. You can't attack Ruins, occupied or not.
