My question is if i use shackle soul on an enemy unit and it causes them to take Leadership tests to move, run, shoot, assault, etc. And i assault him in the same turn, does he have to pass a leadership test to swing back at me in the assault?

Nope, because in your example they are not assaulting they are the defenders reacting to your assault so they are not doing one of the specific things you are required to test for.
