My question is, If the tankbusta unit starts the turn in a trukk, do they then have to disembark and attempt to charge to satisfy the part of the rule which states they must always try and assault the nearest vehicle or can they stay in the trukk?

No they are free to move as they like in the movement phase; and since they cannot disembark outside of the movement phase they are not effected by Glory hogs in the assault phase(but assuming they can shoot, they must shoot at the tank)

FAQ supporting text:
Q. Can a unit of Tankbustas that start a turn
within line of sight and/or charge range of a
vehicle, move out of line of sight and/or assault
range in the Movement phase in order to avoid
firing or charging the vehicle?
A. Tankbustas are free to move as they please in
the Movement phase. They are only bound by the
‘Glory Hog’ rule when shooting and during the
Assault phase.

Tank bustas are only forced to fire at a vehicle if one is visible. No other restriction for movement or assault, though you will rarly be able to assault a regular unit if a vehicle was in sight. They will fire their 24" rokkits even at a basilisk 120" away.

If, for example, they blew up their own trukk blocking LoS to an enemy vehicle by failing the bomb squig roll, they would still not be force to assault the now visible vehicle.
