Now we have been in the the same point in time in 40K for quite some time, my question is where do you want to see it go?
What big event or events would you like to see happen and the following ripple of events to follow?
Ex;
Emperor dies, Imperium disintegrates blah blah so on and so forth.

There's not many directions they can take it without having to remove entire ranges of models from the game. The two outcomes that I could see being good for fluff and the game itself both involve the Emperor.
He 'dies', gets reincarnated, cleans out all the filth on Terra and it's beaurocracy and leads humanity on another great crusade, this time not entrusting the crusade to genetically engineered sons. This, however, is not very grimdark, and is too hopeful.
The other is that the Emperor actually dies. He is fully awakened in the warp and becomes an actual god. The astronomican fails, the Imperium crumbles and smaller Empires are carved out by various human factions, most notably Space Marine chapters. They fight on their own, having to rely on astronimican-less warp travel and generally are dying out. Don't go with the theory of the galaxy becoming a giant Eye of Terror either. That's more grimdark.
Neither of these require much more than a fluff rewrite for the Imperial Guard and Space Marine chapters, and the whole miniature range is still completely viable. The second option also opens a lot of room for people to get creative with their own fluff.

Emperor dies.
Things get really bad with most of the Imperium being taken and with only the heavily fortified worlds or simply the most stubborn holding on.
Chaos takes over aproximately half of the ex-Imperium. Orks gain larger and larger empres. The Tyranids begin to come into contact with Chaos and Orks more than anyne else. Another Necron World Engine awakens but it is destroyed by the Eldar in an attack that claims the lives of many Eldar warriors.
A new figure supposedly the new Emperor of mankind steps forward. Whether he is the Emperor reincarnated or not no one really cares.
The astronomicon restarts and contact is regained with dozens of besieged worlds.
The new Emperor begins to launch a new Great Crusade at the head of a huge army of faithful. As they connect with more and more marin chapters or IG strongholds the army swells and becomes better and better traied and equipped.
It stops there.
Chaos are hte bigger players but are beset by the Tyranids.
Orks are gaining strength but can't quite overcome the Tyranids.
Necrons are beggining to wke up in force but are being halted by the Eldar who pay in their own blood.
The Imperium is becoming more stable but still has a long way to go.
I'd like to see that...

I'd simply like to see the outcome of the major campaigns and battles GW started.
Like finally knowing the real outcome of the 13th Black Crusade on a much larger scale. Will Chaos hold on to the worlds it conquered, or will it be pushed back into the Eye?
Will Helbrecht and Yarrick finally capture Ghazzy?
What will happen to the BA? Will they defeat the tyranids converging on their homeworld? Will they survive the onslaught of Ka'Bandha?
What will happen to the Raven Guard, facing the full might of an ork Waaagh! after their failed raid on Kastorel Novem?
How will the Tau Third Sphere of Expansion end?
Will the Imperium finally defeat Leviathan and all its different splinter fleets?

My guess? Some cash-in campaign involving whatever the newest army is as the protagonists, probably involving a forgeworld somewhere.

I think it may progress into a massive Black Crusade, That will then be BEATEN. Then it'll start all over again in the 42nd millennium.
And the Emperor will never die, because that would kill 40k.

Add some new stuff, kill some old stuff, nothing too drastic.

I would have loved to see some small portion of mankind led toward some technological enlightenment. Let some world somewhere have a completely functioning STC library.
Of course, this would completely upset the whole grimdark thing, but a glimmer of hope for mankind showing some of the strengths of the past would be nifty.
That, or just let the Space Termites eat it all and close the curtain.

Would be nice to see the Necrons awakening fully.
The imperium would face a threat rivaling a tyranid hive fleet, and the adeptus mechanicus would most likely be split down the middle creating some chaos at the heart of the imperium.
Chaos would be faced with legions of soulless incorruptible foes.
The eldar lacking the Blackstone fortresses would have to cooperate/manipulate an IoM in turmoil.
The orks would have the time of their lives, would love to see what the mek boyz dreamed up.
Tau would most likely try to move in on the imperial worlds near them.
Dark Eldar would most likely not care much unless the necrons threatened the webway.
Tyranids would be faced with a foe capable of greatly diminishing the biomass of a hive fleet with each engagement, as there would be nothing left to devour even if the nids won.
Would drastically change the galaxy without things getting less grimdark, and units disappearing (adeptus mechanicus would most likely be worst hit, but not bad enough to warrant removing units from the game.
The warp could be in some danger, but not immediately.

I think a good way to advance the story would be for Abaddon to break out of the Eye of Terror (finally) and go on a rampage across the galaxy. GW wouldn't have to discontinue any models; in fact, it provides an opportunity to create more units for Chaos!

-Cadian gate is broken allowing Chaos to gain a foothold in real-space. -Imperium is forced to reinforce this area to halt further incursions into Imperium territory. -The lack of support and rescources on many other fronts allows other races more freedom to expand and prosper (Tau). -Ork's seeing lees of a fight in the Imperium defending against them concentrate more on the 'nids being that is where the fighting is. This also triggers a reaction from the hivemind to create yet more ferocious creatures and develop new tactics. -Eldar go on as usual for the most part, striking chaos in tacticaly sound points such as supply centers and other vulnerable locations. -Tau gain more followers when Imperials hear about the advancement of Chaos causing fear and lack of faith in the Imperium, therefore many begin to follow the cause of the greater good. -Dark Eldar follow the tactics of the Eldar and strike vulnerable spots torn by war and disease from the new Chaos crusade. -Increased fighting obviously spreads to differnt sectors of space, encounters with Necrons increase with the fighting taking place on tomb worlds more often than usual. -Space Marines are more eager to fight than ever, the ultimate disgrace to the Imperium now a very large threat. That is my vague description on what I would like to see
