As in, how does it have such a huge explosion when detonated? Is it ever explained?
I can think of only 2 explanations:
-nuclear fission
-E=mc^2

Or am I delving too far into the fluff

Presumably a very effective nuclear reaction.
No one ever said it was simple or cheap, could well be (and probably is) a massive use of resources.

Not far enough, actually. Eterminatus is not 'blowing up a planet.' It's one of several things that absolutely wrecks a planet, like virus bombardment which rots all organic matter down and then ignites the flammeable byproducts of decomposition, devouring everything it reaches and then incinerating the rest. Or burrowing cyclonic torpedoes, which dig deep into the interior of a planet before detonating, resulting in massive tectonic instability, and may actually rip it apart if everything goes perfectly.

Well thanks for that, I never actually knew how Exterminatius worked. all I knew was from DoW2

DOW2 is a good overlay of the Exterminatus process, although you need to know the process to understand it fully.

an Exterminatus is typically started off with a conventional bombardment to destroy any bunkers that could allow for survival of the Exterminatus proper.

Then you will use the Virus Bombs. The virus destroys all living material and causes it to break down, essentially a rapid decaying process. This causes a massive build up of flammable gas in the atmosphere.(this isn't really shown in the Dow2 movie)

Then you will fire a lance or explosive ordinance shell at the planet. This is to set the gasses released by the Virus on fire.

The ensuing firestorm will incinerate anyone who managed to survive inside a sealed suit but is still on the surface(this is what the last bit of the Dow2 movie was)

Finally, you have the option of actually destroying the planet itself with Clyclonic Torpedos. These burrow down into the planet's core and detonate their payloads to blow the planet apart. the result is a new asteroid field.

Is the final shell shot out in DoW2 an incendiary round or Cyclonic Torpedo?

That would be an incendiary round. Torpedos are, well, Torpedos.
