So I'm trying to decide which one my seerstaff archmage will take. I am already taking dwellers, regrowth, and throne of vines.
But I'm having a tough time choosing between 4+ regen and +4T.
I suppose I could have my lv2 take earthblood and just have it be a 5+ regen as well.
Any thoughts?

I would take both and not take Dwellers at all.

so play completely defense? I'd like to actually kill some things with magic instead of just making elves that don't die.

I believe that defensive magic is more reliable and more effective. If he can't kill your dudes as effectivly, but you can still kill him just as reliably as before then you have an advantage.
This is especially true with the infantry vs infantry grind battles. It isn't the deadliest infantry that win, its the ones that die the slowest. This is also reinforced by needing to kill a unit fully to gain the victory points.
Dwellers is good, but I prefer the defensive combination of +4 toughness and regeneration. If you are really tough AND have a good save you increase your survivability by a huge amount, especially if you are facing lots of Str3. Str3 wounds T5 the same as T7. Regeneration would make it even more effective.

I think it's psychological too, people are more likely to throw all their dispel dice or a dispell scroll at that big spell that has the potential to kill a lot of models rather then the little spells that will allow them to win combat. For my Gor if I had the option of losing 1/3 of my unit or making it so I wound you on 6+ and not 3+ on the 30 attacks (40+rerolls roughly 30) I will hit you with I'd take the dwellers.

Thus, After having thus successively taken each member of the community in its powerful grasp and fashioned him at will, the supreme power then extends its arm over the whole community. It covers the surface of society with a network of small complicated rules, minute and uniform, through which the most original minds and the most energetic characters cannot penetrate, to rise above the crowd. The will of man is not shattered, but softened, bent, and guided; men are seldom forced by it to act, but they are constantly restrained from acting. Such a power does not destroy, but it prevents existence; it does not tyrannize, but it compresses, enervates, extinguishes, and stupefies a people, till each nation is reduced to nothing better than a flock of timid and industrious animals, of which the government is the shepherd

Maybe you would consider not taking Throne of Vines.
having +2 toughness and 5+ regeneration on a unit is better then +4 toughness or 4+ regen by itself.

a level 2 has only a 1/3 chance of Shield of Saphry(unless its the Sig spell?)

As has been said, Shield of Saphery is the signature spell, otherwise it'd be hard to pick it on a L2 without Seerstaff.
