start with the tau, because it would be easy.
but there is a serious issue with fighting a war on all fronts, placing all the focus on one objective will make you lose all of the others.
see WWII and germany.
the other problems is that once an empire becomes too big it starts to collapse in on itself..
see Romans, Greeks, British etc.

Tau would go first if they were targeted, though they probably won't be. Maybe containing the 14th black crusade, since it's broken out of the Cadian defense.

Well I'm not sure about all of the Imperium, but the Spacemarine chapters should really try to focus and work with eachother better than they currently do. They should focus on the Tyranid threat first and foremost I would think.
Of course that doesn't fit in with GW's fluff. They want the Imperium and the SM chapters to be spread thin. The whole ... dark grim thingy.
edit: Also, GW doesn't want any of the races to be squashed by the Imperium. They want to sell models!

1 on 1 Imperium could beat anybody, no contest.
The only real problem it would have with Orks, Tyranids and Chaos ( both Eldar are to few, Tau are to small to be able to resist and Necrons are not united ).
The problem would be defeating Chaos Gods ( who could always corrupt naive Humans ) and Orks ( who are so stretch that it is funny to even consider hunting them all down ).

Romans fell because of the barbarians and inner fight.
Greeks fell because of the inner fight and foreign invader.
British fell because people of their colonies wanted freedom + that was one of the main acts in US - British contract in 1941 - to dissolve the British Empire.
Imperium only problem would be inner fight that would be resolved by massive invasion fleet.
And there are other galaxies to conquer...

Tau eldar. Ya they can beat them one on one everyone lining up to fight kind of thing.
Orks, no. Not a chance. No one has been able to stop the orks. The IoM even tried.
Tyrnids. Maybe they wouldn't look good after the fight no matter how it went.
Chaos. That's impossible. Chaos comes from within. It's like fighting a war with yourself. It should say a lot how chaos was the side that destroyed the empire.
Necrons. Maybe. They might not be united anymore, but they are still bloody powerful. They can blow up stars and one can even time travel.

They should focus on the Tyranids, they're pretty much their biggest threat right now since the Newcrons got retconned.
They'd probably focus on Chaos though. Hell maybe they'd even invade the Eye of Terror. I wouldn't want to be the first wave of Guardsmen in that....

Guys, it says if they were ABLE to focus. Thus, if the Imperium suddenly lost all of its enemies save this one enemy.
I think only Tyranids and Orks would give a real problem. Chaos would be conquered easily, but then again, the real problem with Chaos cannot be solved with a military solution.

The hardest part about defeating Chaos is the same problem America faced in Vietnam and now in Afghanistan.
Chaos is just going to retreat across the border to Pakist- I mean the Eye of Terror, where it's going to be nigh on impossible to root them out of.
The Tau? By all rights, the Tau should have already been crushed a long time ago. Their plot armor is thicker than anyone's in the game, lol. The Tyranids? Eh, it's been done at least twice. The problem is chasing them off into deep space to find where they come from. Orks? Probably like roaches or ants. Can probably drive them out of your house for a little while, but there's always a few you missed. Eldar? Definitely. If a single chapter can wreck a small craft world, the combined weight of all of them should be able to do it in short order.

Actually with the combined might of the Imperium and the psychic power of all the Imperium's psykers, and the power of the various saints and living saints and chapters of the astartes and so on and so forth, the Imperium could conceivably do a great deal of harm to Chaos even within the Eye of Terror, perhaps even gaining a permanent foothold and beginning to literally cleanse out the Eye, using it as a pocket of warpspace that is strictly under the control of the Imperium.
But even then, even after wiping out the entirety of the influence of Chaos within the Eye like that, the problem is that Chaos is not a physical enemy from without. It is an enemy from within.

The gods seem to get weaker the less people exemplify their traits. For example the less disease there is, the weaker Nurgle gets. This may serve to the Imperium's advantage as the Gods' followers are killed off.

Yes, but Humanity is a violent creature at heart, and so its very existence feeds into Khorne. People will always seek greater pleasures, or seek to achieve perfection in a given area, whether that's beauty, athleticism, art or whatever. All of these things feed Slaanesh. People always desire change. Change is the only constant in the universe. Well, that goes to Tzeentch. And disease? For as long as there have been organisms, there have been bacteria. Bacteria cause disease, virii cause disease. There will always be disease, death and decay, so there will always be Nurgle.
