Would there be a profound shockwave effect in the warp if he died? would it cause a surge in power making all psykers go nuts? would it kill them? what would happen?

Nothing would happen to them. It's not like his psychic presence has any form of control over every human psyker in the galaxy....

Well, if Big E is really Warp God in making, then there'll be ripples on scale of The Fall, with innumerable victims(hopefully not creating new EoT but something like High Haven). But also there'll be steep increase of psyker emergences, Sanctioned Psykers will be lifted to next level and weaker ones wil be able to survive.
If Emperor is NOT a God, then i expect that all Sanctioned/Soulbund Psykers will be in for the harsh ride, with most of them dying, either instantly from backlash/loss of their focus or later when there'll be demonic feeding frenzy.
PS Just occured to me that while Slaaneh birth created EoT question remains where is "cradles" of other Ruinous Powers? Maelstrom is big enough to be one, but the other two?

Most psykers would likely feel it, aside from those who are beyond the astronomican. But I don't think it'd kill any that were outside of the solar system.

Hasnt happened yet. Time is nothing in the warp no logic behind it. Look at the great psycher armys one will bear one at its peak and another the other at their peak. Then mass death possibly even eraddication if not prepared fo war with a chaos god.

I kinda think it will affect the human psykers badly since most of them are not fighters and simply connectors exchanging messages. While the heretical ones are not nearly connected so they would not feel it as much. i think most loyalist pyskers would die

The Ork wierdboyz draw off the Waaagh field generated by nearby orks, not the warp directly IIRC.
As to the rest... depends on what propaganda/fluff you favor 
The SW claim that rune magic isn't Psyker, so that could be an interesting acid test of thier claim.. Automatically Appended Next Post: NId Pyskers are channeling the Hive mind, not the warp, IIRC. Could be wrong on that, as it's been a while since I had Nids.

Well there should be a lot more of them, for starters. But if he died, according to the Imperial propaganda that is the background, the Astronomican would collapse.
And how isn't the Emperor not a god? He was born (allegedly) in 8000 BC!
