When your squad gets challenged during Assault, do you automatically challenge any character/HQ choices in the squad? or can you choose who accepts the challenge?
For example:
Orkz Boyz Mob
w/ Nob
w/ Big Mek

I get challenged, does the Mek HAVE to fight or can the Nob take his place since he's a "Sergeant"???

Page 64 of the basic rule book holds the answer you are seeking. The section is titled Accepting A Challenge and outlines how you go about doing just that, how you go about figuring out which model in question is the one you need to select. There is even a bold section which will draw your eye, and you are in luck that it is the very answer you are seeking. The rest of this section will go on to better explain what this one line means, making it very clear how you go about figuring out whom accepts the challenge.

This is true, but:
Remember that a Character can only accept/Issue a challenge if he is engaged in the combat. I.E. in Base contact with an enemy unit, or within two inches of a model in the same unit that is is base contact with an enemy unit.

Deathreaper, you have just made my life a little bit better. If a challenge is issued to a bunch of boys with a nob out of combat, then the nob cannot accept, yes? I'm just making sure I interpreted that correctly.

Also, if there is no challenge accepted, can one be later issued once the nob is in combat?

Well of course prior to the I10 step, as that is when Challenges are issued
