How do you remove your mold lines? Do you do it piece by piece or glue the whole model together before you scrape them off?

Piece by piece for three reasons.
- Firstly it's easier to clean them when you have full access to all angles of the piece.
- Secondly if you assemble the model and a mold line is in an area with a weak joint chances are that model's gonna be in bits anyway.
- Thirdly if you assemble the model and no joints are weak then I guarantee you'll see a mold line that is clearly visible but inaccessible.
Best advice is to do the mold line cleaning whilst watching TV, makes the tedium less so ^^

For the reasons expounded above.
To be frank it has never occurred to me to do it the other way.

Piece by piece. The only exception is if I find a mold line that I somehow missed during prep (after priming, inevitably). If it's glaring enough, I'll shave it down and touch up the area with acrylic before I move on to actual painting. I try to be thorough, though, as I find cleaning mold lines on an assembled model awkward.

Piece by piece; and its a pita!!
But the difference it makes is huge, so its gota be done :(
Sometimes clean a piece then glue then clean the next ... etc.
sometimes sit and watch a movie and clean all required bits in one go.
- either way i seem to end up covered in grey styrene shavings! hehe

Lots of great advice in this thread - I can second almost all of it.
I do min piece by piece before assembly, and will fill seams and such post assembly.

Always piece by piece.
This is definately something you want an audio book, podcast, movie, bunch of friends to chat with over for.
I usually wash the pieces/sprues, clean off mold lines, assemble (depending on model), convert/GS, wash again then prime.
