If I were to cast an upkeep spell (not upkeeping a spell already cast, but cast it for the 1st time in the game) will my opponents spell to remove all upkeep spells remove this spell? I ask because I was wondering if a spell is considered an upkeep spell until it is actually up-kept. I am assuming it is, but it was an interesting thought that was brought up.

Yes. An Upkeep spell is an Upkeep spell even if you never pay to upkeep it.

He can definitely cast Purification the turn after you put it into play.
