Looking at the Inquisition codex, it's quite clear that it's designed to be more of a supplement, plugged into other armies as support units to bring boosters to Space Marines, IG, etc.
However, if one were so inclined, would it be possible to create an army where the main force is Inquisition and isbacked up by smaller allied forces, such as a couple Knights, or a small Skitarii Maniple?
Obviously they're not flush for choices, and you'll have to max out on slots and Dedicated Transports to start even reaching a decent points amount, but do you think it could be done?

I've seen a couple of solid 1500 point lists of pure Inqusition.
Usually they have:
- Coteaz
- Generic Inquisitor
- Large Henchmen Squad with Servitors and Jokaero Weaponsmiths.
- Couple of Henchmen Squads with few Acolytes, maybe a single Psyker for Warp Charges. All inside of a Razorback with Psybolt ammunition.
- Maybe a Land Raider filled with Crusaders and Death Cult Assassins.
Note that you may technically have infinite Inquisition Detachments, so if you have three HQ's you can have up to nine Elites.

While you would need multiple HQs and it could get repetitive, Inquisition do have some very solid units to build around.
For shooting, you have your basic cheap-as-chips Bolter Acolyte squad, who can also be given Storm Bolters to hang out at longer range without losing firepower, or can take up to 3 Special Weapons per squad (and can also be used in Chimera). You then have the option to add in Heavy Weapon Servitors or Jokearo for some real punch and even a 10-point Psyker in each for an extra Warp Charge and potential buff or Witchfire attack. 3+ of these units in some fortifications can offer a pretty decent firebase, especially if you are open to throwing in some Skitarii or Militarum Tempestus as well.
In combat, again you can take cheap suicide/meatshield squads of laspistol/ccw acolytes (3 attacks on the charge at 4ppm is all right, especially if you have the slots to spam them. 120 points for 3 10-man squads that can tie up units for a turn or two makes a nice first line of defence). Then you have the real powerhouses, the feared Death Cult Assassin/Crusader squad. The DCA hit with an insane number of high-init power sword attacks, while the Crusaders tank up front with their 3++ saves. With a Priest to make them Fearless and confer Hatred and War Hymns, and an Ordo Xenos Inq with Rad/Psycho grenades, these guys can handle a anything not in 2+ armour. Buy them a Land Raider or just run multiples and hide them for countercharge duty and they'll clean up!
The only real weakness is anti-air, which Skitarii have several good answers to.

Agreed with both the previous posters...The LRC with Psybolt, Crusader, DCA, Arco-Flagellant, melta-bomb Priest, and Psyker Close Combat unit makes up for one high damage / high durability deathstar...but at 450+ points, it is not cheap. I've run two of those and a Plas-Servitor, Jokaero, Crusader squad in the backfield with a Sentinels of Terra Allied Detachment in some larger tournaments, and while I never came out close to the top, I also never came out on the bottom. The Inquisition is ultimately customizable to do everything, and do it WELL, but it is expensive. You can go on the cheap with MSU units IF your environment allows unlimited detachments, because with only three elites and two HQs (and technically one FA with FW Valkyrie) you are not going to pull it off in one Inquisitorial Detachment.
