This was something my son and I were fiddling with in-between game. Our FLGS had a city mat with several roads marked on it, and it just screamed for doing a tower defense style game (it was the F.A.T mat, hi-tech city 6'x4').
I don't have any solid rules for this, but I'd certainly like to throw out what we did...
1) We set up a central objective to be destoyed (T6, W10)
2) The defender started with two squads (about 100 pts. Each) placed wherever they wanted to defend the objective. Only melee troops can move, and no more that 6" from their starting position. All of the defender's units gain Precise attacks. Those units already with Precise attacks get Precise attacks on any hits.
3) The attacker started with about 500 pts in various groups worth from 50-150 points. The attacker units had to be brought in one squad at a time and must remain in the movement lanes, except when assaulting the central objective. The attacker MUST move towards the central objective if it is able and must fire on the objective in preference to defending units. A new attacking squad cannot be brought in until the previous unit suffers at least one unsaved wound.
Every time the attacker brings on a new squad, the defender can deep strike in an additional unit, reinforce an existing squad back to full health or upgrade an existing squad (replacing the squad with a unit worth 150% the value of the current squad)
Every time the attacker destroys a squad, it can replace or gain a new unit worth 50% of the point value of the destroyed unit. If the attacker causes a wound to the objective, they get 50 points toward replacing lost units (in reserves) or putting a new unit in reserve.
Game ends when either the attacker runs out of units (defender wins) or the central objective is destroyed.
In the game we played, my son was defending with Necrons while I attacked with 'Nids. I brought out, in order, hormaguants, termigants, tyranid warrior, tyranid warrior + termigants, 3 tyranid warriors.
I'd certainly welcome anyone adding to or proposing modifications to these rules, or even giving them a go.
