Can a Hierophant(or any gargantuan creature) gain shroud from a venomthrope? Can they gain a cover save at all?

Why wouldn't they? Doesn't say GMCs can't take cover saves in the rules

Yes, you can. It doesn't really make sense, since per fluff the venom is emitting a spore cloud that provides cover. Which wouldn't even cover half of one leg of a bio-titan. But RAW, it works.

I am fairly certain hierophants can produce their own shrouded with an upgrade. Not 100% on that but almost.

Nope.
What you might be thinking of is giving the heirophant the upgrade that adds a transport capacity, and putting a venomthrope/malanthrope inside.
This used to let you measure the shrouded aura from the heirophants base. However I think the FAQ changed it so aura effects can't be used by embarked models, so it no longer works.
