In a mixed army in which a non-Custodes model is the Warlord and gets slain, can you use 'Shoulder the Mantle' to then make a Shield Capt the WL instead.

I don't see any reason that requires the initial WL to be a Custodes.
The strat becomes available once your Warlord is slain. Full stop. Not Adeptus Custodes WL, not Shield Captain WL, just Warlord.

The only thing required is that the new WL be a Shield Captain.
So could I have a DW Watch Master as my WL, and if killed, spend 1CP to make one of my allied Shield Caps the new WL?

As long as you have access to Adeptus Custard Cream stratagems by having a qualifying detachment, yes, you may. Like you said, the only requirement is that your Warlord is slain and the new Warlord is a SHIELD-CAPTAIN, not that the slain Warlord is also a Delicious Custard Cream.

That's pretty gross. So with a Command detachment of Shield Captains, and 1CP each time the "WL" dies, you could ensure your opponent never gets Slain the Warlord unless they REALLY dedicate to killing 4 Character models (the initial WL and the 3 Shield Caps).

I very much see this as being the fluff intention of the strat.

Like there's a Guard commander in charge of the battlefield (which generally isn't a Custodian's job anyway). But the leader dies, and so the Shield Captain steps in to take control.

I mean, you can't if your opponent kills both in the same phase, since you can't in Matched Play (aka the Default Play Mode for 99% of people) use the same stratagem twice.

Oh certainly, but if you have 3 Shield Caps, don't pick the one that is in a situation that could be killed in the same phase as your current WL. Killing Characters reliably is very rare, so it should be easy enough to select the right Shield Cap to be the next WL.

And the Strat allows you to pick a new WL trait, so the Cap could get one of the buff traits to help with this
