At least a few of them, yes. Perhaps only in-system but still off their planet.

Guilliman traded with other planets before the coming of the emperor.

Dorn had his space fortress before the coming of the emperor

Alpharus/Omegon led an entire spacefleet before Horus found them.

Corax may have known space.

Your research could use some work.

May I enlighten you to the existence of http://www.lexicanum.com/ and http://warhammer40k.wikia.com/wiki/Warhammer_40k_Wiki?
There you can find the answer to all of the questions you have asked so far.
If you have done 'research' already, it clearly was not enough. The answer to this question can really easily be found there.

Trade aside, Guilliman ruled over Ultramar, which comprises a bunch of sectors.

And Magnus was aware of space and warp, iirc he "met" with the Emperor via warp projections..

At the time of his finding, Jaghatai Khan had just conquered the planet he crashed on, and according to the background in the White Scars Supplement, there is discussion of the fact that he had plans to expand his empire into space. He recognized the Emperor as the person with the power to help him expand into space. That's why Khan swore allegiance to him at all.
