I've read a handful of Space Marine related books that I've really enjoyed. And from reading some wiki type pages over the years, I know the very basics of the Horus Heresy and I know of the Emperor's body surviving in semi-corpse form on the throne.
When I look at the vast amount of books written about the 40k universe, I don't know exactly what to choose to read the actual story of these events. Is there a specific book that covers the Horus Heresy that also covers the Emperor going from a living-breathing person to what he is now? Or is that something that is only covered through a long series of multiple books?

the Horus Heresy is covered in a long series of multiple books called... the Horus Heresy by and large they're pretty good. Inciderntly the series has not yet GOT to the Battle of terra let alone it's end.

I'm aware of the series, I was just hoping that there was a single novel or maybe a couple of books of the core story.

nope. but thats because we've not actually reached that point. I don't expect the seige of terra will be in a single book though I expect GW to stretch it out an insane amount

This is the fluff as it currently stands but is about 25 years old. GW have explicitly stated that they have a different vision for how the battle will play out.
The old Bill King story of the Emperor vs Horus is told from his point of view and makes him seem very human. The Horus Heresy novels have depicted him very differently (with no stories told from his own point of view) so I suspect they will update the story in line with that.

I vaguely remember them saying it was only going to be a 10 or so series book. Look what happened.
At OP currently Sangys just starting to tiki touring his way to Terra. So come back in 10 years and Emps might be in the throne. Might.

Don't spoil things.  Those Bill King stories will always be canon as far as I'm concerned; what do Black Library know?

From what I understand is that someone (extraordinarily physically gifted) needs to be on the throne at all times to protect Terra from demon hordes invading through the web way gate under the palace.
Emperor did it until he absolutely needed to confront Horus on the Vengeful Spirit, during that time Malcador took his place. When the emperor was dragged back to the throne after getting his butt kicked, Malcador collapsed into dust at the same moment that The emperor replaced him.
This of course is the very simplified version of the events.
Once Magnus breached the webway, it allowed countless hordes of demons into the system which had to be repelled by the sisters of silence, the custodians, and various irregulars of the mechanicus. The only thing holding them back at this point is the emperor on the golden throne which also acts as a life support device..
