Just read all the awesome Punisher comic series and am seriously wondering if the Punisher could take down a space marine in one on one combat? keeping in mind what i've just read was him killing every superhero out there!

Now don't get me wrong, I actually love the Punisher. He's one of my favorite Marvel character, but he's at his best in his comic where he doesn't deal with all the super-powered bullcrap of the marvel universe. The Punisher Kills the Marvel Universe is not a 'great' comic  . It's actually fairly terrible and was meant as a joke even when it was written.
Several of the super heroes and super villains we see the Punisher kill have survived much worse than what he does to them in their own comics...
Venom is all but immune to electricity, it's been shown to have no effect on him and other symbiotes several times. Not to mention that killing Brock would not kill the symbiot.
To think that Dr. Doom's armor isn't shielded against EMP when run of the mill military tanks are is flat out slowed. And a sledge hammer would break off long before Doom's armor even dent. I mean come on, that armor has taken full strength punch from the Hulk and held up... Power Armor has nothing on Doom's armor.
The scene with the mutants is laughingly bad... as if you could just 'send a challenge' to people like Magneto, Apocalypse or Dr. Sinister and they'd come running. What is this? The old west?  Not to mention that Magneto alone has been known to detect the signature of incoming warheads and neutralize them before they ever became a problem at least twice in other comics...
To think that Castle could outfight Wolverine in close combat is also fairly silly - I know the idea is that Wolverine is drunk, and thus not at 100%... but you see, his healing factor prevents him from getting drunk in the first place... so der? Wolverine himself has survived much much worse than the electricity he receives at the end of their fight - hell, as much as I don't like Wolverine, he's been shown to be practically immortal at this point, odds are the generator would overload before he died.
Oh yeah, and Captain America would turn his back on an enemy. In the middle of a fight. Right.
Against Daredevil? Castle is a man... he takes a beating and gets thrown off a 5 story building and than still has the energy to stand up and stab someone? Heck, he'd be dead or in the hospital for months...
In short, for the story to work, the entire Marvel Universe was 'pussyfied' to the nth degree while Castle himself was 'boosted'. That is not to say Castle couldn't kill many superheroes (In Welcome Back Frank, he takes apart Daredevil in a really smart way and could easily have put a bullet in him had he wanted to), but the ways they show in that story simply wouldn't work - and I think that was on purpose, to show us it's a joke.
So than, the real question in the Punisher vs a Space Marine is, those the author have the permission to make the Space Marine into little bitches like Ennis was given with the Marvel Universe? If not, Castle is done for. In his own comics, he has a hard enough time dealing with normal (See Barracuda or The Russian, pre- op), non-super powered baddies.

 Ah Tarkand, it's good to see someone else reading these as well! I did get that majority of the super heroes were slightly dumbed down but never the less entertaining for what it's worth. the russian would have to be the scariest bad guy he faces by far! Barracuda was an animal but that russian was a nut job! kinda like a space marine with breast implants! i'd like to think castle could at least have some chance against a space marine? but even that gang of mob widows nearly capped him? so i suppose on a good day he could deal the pain to a marine or on a bad day he can get snipped by a widow with a shotty! I mean he can't be so totally awesome every single day can he? (please Frank if you read that....kidding)

One of the thing I personally dislike about The Punisher is that they suddenly get beefed up when in books with other Super Heroes. In his own comic, Frank gets captured by a squad of Delta... he manages to get out of it, but the point is, a squad of well equipped, trained and intel'd normal human are able to bring him down. But when he shows up in Spider-Man (or whatever), all of a sudden he can keep up with mutants, genetic enhanced human, demi-god and what not. Same goes for Batman who has to exert himself to take down people like the Joker in his own comic, but can keep up with Superman and Captain Marvel when it's a JLA comic... it's all very silly, I mean Batman would die from the colleteral damage of some of those JLA fight.
In short, the fact that the widows almost got him doesn't make him any less of a bad ass... I mean, even the world's deadliest man is going to die if he gets shot in the head from 300 yards by someone with a sniper rifle.
As for the actual question - if it isn't bullet proof, Castle always has a chance. We now Marines aren't immune to small arm weapons, so obviously under the right conditions and with the right condition, he could do it (So could anyone really)... but realistically speaking, he'd get destroyed.

Chalk up another thought about Leman Russ Punisher against Space Marine armies. Instead we just get another one of these awful threads. I'll just pretend you posted a legitimate question:
A Leman Russ Punisher might seem pretty nasty at first, but only about 3 of those 20 bullets will get to both hit and wound. That generally means one dead Marine per round of shooting. Taking Pask helps this, but you'd be better off taking a standard Leman Russ Battle Tank, or maybe even a Demolisher.
There, that feels better.

Um, actually, the Joker is never shown as a serious physical threat to Batman, so that comparison wasn't really apt. In addition, you're not really taking into account that Batman has his won super-powered enemies that he has to defeat and thus is at least prepared for the enemies faced by the JLA, whereas the Punisher, to my knowledge, does not.
Anyway, the Space Marine would win. Even Frank Castle couldn't take on a full astartes for long; these guys are capable of punching through tank armour.

In the 40k universe, both on the tabletop and in the fluff, heroes beat regular soldiers.
Human heroes beat regular Space Marines. It has happened, many times. Gaunt, Straken, Schaeffer, Cain, Yarrick - they curbstomp rank-and-file Astartes. The Punisher would have no problem.

Nice use of "etc" there because Gaunt, Schaeffer and Cain don't have special gear and still whomp Space Marines without effort. Also powerklaw is special now, so rare every Ork mob has one.
But you make a good point, a 10,000 year old book written by an incompetent who was tricked over to the ass-end of the galaxy while his leader was killed is invaluable. That's why Ultramarines are such useful soldiers, and totally haven't been absent from every major conflict ever.
Horus Heresy? 13th Black Crusade? 1st, 2nd AND 3rd wars for Armageddon? Pff, the Ultramarines were too busy getting their 1st company ate by 'nids.
