I'm not sure if it's just in the video games or if it's covered in the fluff as well but Inquisitors are massive! Looking at Toth in Dawn of War or Drogan and Thrax in WH40K: Space Marine they're about as big as a normal Adeptus Astartes. Is there a source anyone can direct me to that explains this? Do Inquisitors go through a similar training and surgical process or is it some form of cybernetics? Can space marines become Inquisitors?

Inquisitors come in ALL shapes and sizes. Some are short, some tall, some fat, some skinny. At least one comes in a box (ravenor). Some have power armor, some show up scantily clad. In short, the size/shape or disposition of any particular inquisitor is completely up to the person that made them up. Most likely the ones you've seen have had extensive mechanical augmentation.

The Inquisition is probably one of the most diverse groups in the Imperium, right alongside Rogue Traders. There's really no uniformity among their ranks in terms of appearance or tactics.

In the case of Dawn of War, Toth was wearing power armour so it makes sense he was the same size as Angelos.
In Space Marine, Drogan at least was a head shorter than the Marines, although he was just as broad. The other Inquisitor was only seen at the top of a ramp so we can't tell how tall he was. I think this is a case of applying 40k's "28mm heroic" scale to 3d graphics.

In Space Marine, Titus is explicitly about 7'1".
Comparing them, IIRC that would make Drogan about 6'5" or so. Tall, but not massive.

The only issue I'd have with this is that, even in power armour wouldn't Toth still be smaller? While the power armour augments his strength and such it still has to fit his frame, which is much smaller than a space marines.

The height, the weight etc of the Inquisitors are different as how many world of the Imperium they have to protect.
Like many users have already said there are soooooo many different inquisitor 'cause the physical condition of a subject is of secondary importance.
What matters the most is the loyalty to Big E., the Imperium, and their intelligence.

Um guessing its cos the inquisitors not only get power armour but also probly get fed better than most imperial citizens/soldiers and also get more excersize than the usual upper class types that do eat well

Here's one showing an Inquisitor who's only taller than his compatriots thanks to his comedy hat:

One would expect a martially inclined Inquisitor to be big and strong to start with, and some types of power armor add a lot of bulk for even more impressive size. And ofc, when dealing with marines why not send in an inquisitor who speaks their language (carries huge guns, flexing huge muscles, needlessly smearing himself with oil and so on).

That's not a "comedy hat", that is a "Sacred Hood of Inquisitorial Witch-Findin' Pimp-Stylin'".
In my DH games, if an Interrogator makes it to Inquisitor, they get a rosette, a bolt-pistol and one of those hats.
