Hey there. I play 40k a lot, but I don't know ANYTHING about FB. What are the main differences between these Warhammers? In terms of rules, gameplay, whatever.

Is 'Eavy Metal Calling?

Well, obviously the setting is completely different, so that informs a lot of differences. For example, shooting is largely the focus of 40k, but in WFB it's often just a precursor to CC or for pestering weak units. On the other hand, a well-timed spell in WFB can kill dozens of models, while in 40k Psykers are far more often handing out minor buffs and the odd zap. The model count is also typically higher in WFB, and units exist in large blocks rather than looser squads.

The main things I can think of:
Charging is done in the movement phase, and units still move even on a failed charge.
There is no unit cap. Where an Eldar Guardian squad is 10-20 models, a High Elves Spearmen unit is 10-??? models.
Unit selection is based on percentages. For example, with 40K, you could take a 1 HQ, 2 Troops, and put the rest of the points in Heavy Support. In Fantasy, you must spend a certain amount of your points on Core units (basically Troops), and can only spend a certain amount on Rare and Special units (basically Elites/FA/Heavy Support).
Magic is similar to the Psychic phase, however, it is a bit easier to deny a power.
Units form ranks and files, where as in 40K they tend to spread out a bit due to smaller unit sizes.

Greebo had spent an irritating two minutes in that box. Technically, a cat locked in a box may be alive or it may be dead. You never know until you look. In fact, the mere act of opening the box will determine the state of the cat, although in this case there were three determinate states the cat could be in: these being Alive, Dead, and Bloody Furious.

There are some units that have caps.
Combat is less dependant on wounds dealt as things like banners help you win.

Magic is vastly more important, more powerful, more interactive, and you don't get armies that are stuck with no wizards and no ways to do anything about them the way Necrons and Tau get screwed over by the psychic phase.

Cover affects the 'to hit' roll rather than providing a save
Models can take multiple saves. Eg: Fail armour save then pass a ward/invulnerable save.
