We had ways to resupply between missions. However, it took significant effort.
For Deathwatch, Special Issue Ammo was devastating- and also scarce. And the rarity was also quite high, which meant there was no assurances that you could get more after the mission.
For our Heretic Astartes campaign, you could 'restock' between missions- however, your 'minions' were dispatched to scavenge and purchase bolter ammo. So we had a roll to see what quality it was. "Legion" bolter rounds were your standard, with a 4+ ammo check. However, your minions might be able to get you ammo... but it's reloaded, poor imitation stuff... making the ammo check harder (it's gunked up your bolter, one of the bolter rounds was warped and jammed the chamber, etc.).
What makes it cool: That bolter that splatters everything at first starts to become a bit less of a go-to. Soon, you start seeing more precise, coordinated attacks with minimal expenditures (we had a rule that you didn't have to use the rapid-fire setting, which would make it a bit easier on the ammo check by adding a free roll). Raids on Enforcer precincts start looking more likely. Melee attacks start happening a lot more often, and the Astartes tend to walk away with more scratches and bruises than before when they have to wade into the line of fire and get close.
What makes it hilarious: A World Eater with a Heavy Rock Saw charging an Alpha Legionnaire with two flails... and they both end up killing their own cultists that charge in to help, and they both manage to not only survive... but improve.
