Well, your comparative was "a grimdark Star Trek"... which I feel Firefly matches. However, I agree that a 40K setting should be grimdarker still. Film's like Starship Troopers and Event Horizon can practically be considered live action "visions" of a 40K-like world/universe. But then, it all depends on tone and genre. There's no saying a live action film/tv series can't have a more light-hearted, humorous "vision" for the setting. In which case things like Firefly, or even Red Dwarf, could work just fine.

Give me traditional 2D animation any day, month, year. All these 3D CGI shows that come out now look like puke in comparision. Look at the two Clone Wars shows, for example. Only 2D animation would be able to do 40K justice on a scale level, by comparison CGI to still far, far too limited.

If it were up to me, I'd make it an omnibus of short segments like Heavy Metal. Following an UM regiment for an hour and a half was a drag, but imagine if the focus then shifted to the CSM they were fighting, and then on from there. Everything from punchy action comedy with Orks to brooding horror with Necrons and inspiring heroism with the Guard.
It would make a great intro into the game and its universe if nothing else. That universe is way too massive to cram into a single story

My buddies and I were just talking about this the other day. I think it would be AWESOME to see a good live action movie of the Warhammer 40k universe. Just like ALL of the other blockbuster movies for the past 10 years it'll be an excellent combination of live action and CG (think Harry Potter, Lord of the Rings, ALL of the super hero movies that have been coming out lately, etc). The biggest thing I would like to see though is a bit more than the traditional "Ultramarines Vs. Chaos". Now we all know that the standard Marines Vs. Chaos is the primary driver behind the franchise so you really can't leave it out, but you could do is add to it. I was thinking a cool way to do it would be to do a dual-arcing storyline: Chaos is invading system X because they're all evil and whatnot so it starts out with IG and the local contingents fighting the Chaos and getting their butts kicked till some Space Marines arrive (insert whatever SM chapter); meanwhile in a nearby sector, a different SM chapter (my personal favorite would be Space Wolves  ) is fighting one of the xenos races (Tyranids or Orks). This one is a smaller skirmish battle but the wolves are all pissed off so they follow the orks and engage them in other areas of the sector until we come to find out that the Orks are actually being led/manipulated by a Chaos lord. Basically, two fronts of the same war. Ultimately, the two SM chapters have to work together to defeat the Chaos/xenos invasion. This gives you the ability to stray a bit from the traditional Marines Vs Chaos routine, and you also get to show off multiple races/factions: IG, two SM chapters, Chaos, and a xenos race. But it is ultimately something that people can easily relate to: good vs evil.
When it comes to actors, I just want to see Dennis Storhoi as a Space Wolf. That dude's a bad ass. I don't really think it'd be that great to bring in a buttload of big name actors other than simply to use their names to get butts in the seats. Some though, as have been already mentioned would be great: Ian McKellan is awesome so is Christopher Lee, and the Rock is a freaking beast, he'd make a great Devastator.
Oh, and to whoever said to use Arnold as Marbo......... last time I checked; Sylvester Stallone is the one and only Rambo.

What I think a Warhammer 40,000 movie would need to do is to convey the two aspects of the Warhammer 40,000 universe: the background, and the action. By background, I mean all the ins and outs of the universe conveyed through codices, and the action as in a decent representation of what we're supposed to be imagining when our models are on the tabletop and the dice are being thrown. This is what I think let Ultramarines down - we're just watching a bunch of soldiers in big blue armour shoot stuff and say funny words, there's no life to it. If I were writing a script for a 40k movie (and perhaps you should thank the Emperor that I'm not), it'd be about an Inquisitor whose investigations take the audience from the streets of Terra to the battlefields, and letting us know the significance of it all. And the Inquisitor needn't be the main character either, maybe some Joe Public who the audience can sort of relate to gets swept up into it, much like the role of the companion in Doctor Who.

For me, the suggestion that it be a live action series of Rogue Trader is the winning idea, and what makes the whole idea viable.
You would probably be looking at a series a bit like Battlestar Galactica, as I think that is quite grimdark itself at points. I think you need a big fleet rather than a small single craft like you have in Firefly, as I think you need a blend of internal politics (mutants and heretics) external threats (Orks and Eldar) and external diplomacy (other Imperial agencies). There is a fluff precedent of marine detachments being assigned to Rogue Traders, so you can have marines involved, but not central to every storyline. You can have an attached Cadian regiment. You can have ecclesiarchy, a navigator, the Rogue Trader and his immediate cronies. You can have Inquisitors passing though, new commissars assigned to the guard regiments, encounter the chapter master of the marine contingent, and so on...
Then, you can investigate the emergence of the Necrons, with the Eldar sticking their beaks in. A super-violent but cunning emergent Waaargh, Tau undermining Imperial authority in the area, and of course, Chaos worship within and Chaos violence without. You could get a good few series out of it...

Making a movie focused on space marines would be terrible, it is hard to come up with a character that is more boring than a space marine.

Indeed, which is why I think a normal person sucked up into an Inquisitorial retinue (i.e. Alizabeth Bequin from Eisenhorn if you can look past the whole blank thing) would be a good character for an audience unfamiliar with 40k. Heck, with the varying levels of technology and cultures from world to world in 40k, they needn't even live in a society too sci-fi to begin with.

Meh, if they were going to make a live-action 40K movie, they'd be better doing something in the style of Eisenhorn/Shira Culpurnia, and doing it largely with unknown actors. The setting itself is the "star" of 40K, the characters are just a vehicle by which we can experience it, making some huge bombastic action shlockfest crammed full of well-known actors would result in a gigantic flop.
I love the idea of a TV series following a Rogue Trader dynasty, although to do it justice it would need to be done in a similar format to Game of Thrones; long episodes with really high production values. Better they spend huge sums of money on that than getting "big names" that often just play the same character over and over again with a different name.
