Is a romance sub-plot really all that terrible a prospect? There's romance sub-plots in the Inquisitor series, Cain is a womanizer, and for kick-ass sci-fi movies with romance sub-plots? Well, Pitch Black did pretty good.

I think a live action 40k movie could work. You get CS Goto to write the script and direct it so that his life-long dream may finally be realized. You make it about the 3rd war for Armageddon and cast George Clooney for the role of Helbrecht, Matt Damon as Grimaldus, Danny DeVito as Yarrick, and Meryl Streep as Helbrecht's love interest as a part of Goto's classic fluff butchery. Then just get Linkin Park to write the soundtrack and you're all set.

Depends on the subplot.
It's a 40K movie. It's going to be about Space Marines. There's no reason to option the license if you're not going to use Space Marines. The setting is pretty generic without Space Marines (it's fairly generic with them, but you don't have to pay for the rights to regular guys in space ships and guns).
Though I doubt that was the reasoning behind there never being a movie. It's just not a very strong setting for storytelling. Space Marines are awful characters. They're bland, boring, heavily indoctrinated, quasi-fanatical warriors who spend their entire lives preparing for, and going to, war. And when they do go to war, they fight against cartoon characters.
If the audience is already "bought in" to the setting, like those who buy the 40K novels are, then it doesn't matter. But we're talking about having to make a story that appeals to a wide enough audience to justify the enormous expense of having to create special effects for an A-list, or even B-list, 40K film. It's the same reason Sam Raimi was strongly opposed to using the Venom character in his Spiderman movies (and we all saw how well it worked out when Marvel forced him to include Gwen Stacy and the Black Costume in Spiderman 3). Some characters only work well if you don't think too hard about them.

I enjoy finding the 40k movies in the NOT 40k movies..
Alien..Aliens (forget the rest)
Dredd
Event Horizon.
Blade Runner
Battle los Angeles
District 9
Mutant chronicles ( shame they got a almost decent movie, and 40k still has not..ahh well  )
And many others, I find a bit of 40k inspiration in movies all the time, but maybe one day they will make a big budget 40k movie, since weirder things have happened...
( you will note I did not cite starship troopers as a inspirational 40k ish movie...due to how much I loved the book..and the movie made me very sad. :( )
At any rate I agree any 40k movie will have to feature marines in it, but calling them Astartes may sell better, a bit more exotic, plus showing the creation on them, organs and training would make for a cool credits background for the opening scroll.
