The greatest threat to the Empire is the death (mortality) of the Emperor (without an Heir).

This innevitability will eventually shatter the Eclysiarchy, and with it, the centralized government on Terra.  As the myth of the God Emperor is finally disproved by his mortal demise.  The strongest power bases after the rift will be the Fortress monastery worlds of the Adeptus Astartes Chapters, the Fabricator General and the Martian production world in the Terra system and the Centralized autonomies of Cadia and the surrounding worlds.  But with warp navigation immpossible in the light of the extinguished Astronomicon humanity would be isolated and ultimately doomed.

With the fragmentation of the Empire after the Emperor's demise, the coordinated, centralized effort to defend humanity will fail and the rogue missconnected power bases may not be able to guard the Imperiums old boundaries anymore meaning endless slaughter on the frontier worlds and a massive incursion in once Imperial space by all manner of foes.

Long live the Emperor!

However, a succesor must come forth before time is too late!

Listen, it's obvious.
The greatest threat to the Imperium of Mankind is Mnkinds natural enemy, the one thing he cannot survive witout and yet the source of all his woes.

Wo-Mankind.
Yup, I said it. The 50% of the imperial population (Actually, probably more considering the constant state of war would be working attrition on those men of fighting age something fierce) that has one more X chromosome than is good for it will be it's downfall.
They're already amassing a power armoured legion of Supernuns. Next, a carefully hidden program to genetically modify the Supernuns into 8' tall Space Women. These will then walk around near the Space Marines, fluttering genetically enhanced eyelashes and wiggling genetically perfected bums. The Space Marines will be unable to resist. Lured by their charms, they will form "Stable and loving relationships". The second phase will see more and more chapters refusing to fight because "The wife'd kill me. Maybe next week" or because the war falls on "Our night".
Slowly but surely, the Adeptus Astartes will atrophy and die. Then, the gaurdsmen will notice that they are the only ones not getting any "action" and will revolt, before being snared by the very same trap. 'Nids will be too embarrassed to attack for fears of either screaming women with newspapers flushing them down giant sinks or being captured and made wear cute outfits.
Mark my words. The Age of Man is ending. The Age of Settling Down and Doing What the Missus Says is about to begin.
These are the End Times.

Nids without a doubt. For all we know, they have conquered every single other galaxy already. The fact that they have attacked form two two different galatic edges implies that they have a significant presence outside of the 40K galaxy. Most fluff says that those Hive Fleets already encountered are more like scouting parties...if so, the Imperium is royally boned when the actual fleets arrive.

I think the guys at GW made a cracking job of giving every army a reason as to why they can't win.
Orks, can never agree
Elder, to few to mount a war
Dark Elder, same
Tau, isolated
Marines, not numorous enough
Choas, same
Necrons, out the game really (personal view only)

The only contenders really are Nids and Imperial Guard

of course Nids have to battle through every race to win
Imperial Guard are stretched to far to be able to mount an offencive

So I'm going with Nids at the mo, but IG would win if they were able to bring their numbers to bear.

sorry if I missed an armies out, not intentional

I sometimes imagine that something like this would happen in real life. Given that males are in fact aborted females, perhaps one day humans will evolve to have just one sex, who fulfill different sexual roles depending on their age. With no more men as such, there'd probably be no war at all. *sigh*

Ok, I'm suffering from sleep deprivation atm, that's my excuse.

Men aren't aborted females.
Humans have two sex detemining chromosomes. X and Y. Women have two X, men have an X and a Y. So men actually have all the genetic information to make a female, plus an extra bit.
Anyway, loads of wars have been started by women.
Margaret Thatcher and Queen Victoria are two examples that spring to my mind.

Queen Victoria made very few decisions, she just did what Parliament told her for the most part. And Margret Thatcher doesn't really count as a woman.

My reference to abortions was a nod to Valerie Solanas (crazy feminist who shot and technically killed Andy Warhol, and later becaem a prostitute to fund a drug habit). I'll steer clear of obscure references in the future.

On a more serious note, the fluff shows that each hivefleet is over double in size from the previous excursion. Every time they come, the empire has to throw the kitchen sink to get them to go away, but it doesnt seem to be working. I would not be suprised if GW uses this fact as a premise for an upcoming campaign year as everyone bands together to wipe out the threat...The more they kill, the more their army grows...

Mi.

The empire. As for nid they may spread all over but its nothing some good rad and virus bombs cannot take care of. When in doubt nuke um from orbit, Its the only way to be sure. .Tau are going to be a while there to small. Chaos cant keep there guys from killing each other, Nid and cron both are strong contenders, eldar already had there day in the sun, dark eldar just not enough of them.  So i vote the empire themself there so much like our present day politics it cant help but make them crumble.  That would bring about a a true rogue trader game to see the empire fall. All sorts of mixed up fallen marines, imp gaurd units, mercenaries, it would be great.

The only easy day was yesterday.

Tyranids eat the Galaxy. That's why there's no records after the year 40,000. If anyone else won there'd still be some sort of record of things. Necrons are still around but just sleeping. They don't care if 'nids eat the galaxy and move onto the next. That's what they want anyway.

The empire has tried putting the Tau "in their place"; it hasn't worked yet.

More to the point of the topic: 'Nids, nom nom nom...

There are events in the 42 millennium. Read Cain novels. Anyway...

The last Cain novel chronologically takes place during a Black Crusade with Cain as an old man. That's the only one I believe is set in the 42nd millenium.

Funny how things never change.

Anyway, Tyranids omnomnom everything, Crons have a nice sleep. The only way I could see it turning otherwise is a united Ork WAUGH, the Starchild, or an interspecies alliance (Unlikely, but could happen between Tau and the Imperium, and possibly Imperium and Eldar; however, the tech of the two lesser species has to be spread throughout the Imperium, as it's the only way they'll be able to stand a fighting chance).

Yes I looked it up on Lexicanum. The last date is 41,127. Which makes me think the Cain novels aren't very Cannon. But still: Galaxy survives 127 years after M41 then gets eaten by tyranids, Necrons party. Doesn't really matter at that point.

The greatest threat to the imperium is
+1 to itself. It's stagnation keeps it treading water and prevents the GE's forces from ending one war front and moving on to the next.
