First used in TMP when the Klingon ships approach V'ger and then get exploderised and used in EVERY appearance of the Klingons since, up to and including Disco (slowed down, sped up, it's still there.).

I had forgotten how awesome the music from Star Trek: Legacy was. The gameplay was kinda meh, but the soundtrack absolutely slapped!
Honestly all of the Trek series have had great themes for their time, with the possible exception of Enterprise (good song, but doesn't really go with Trek that well). I really like Voyager's theme myself.
For the original cast's movies, I'd have to go with Horner's score from Wrath of Khan. For the TNG movies, I'd have to say First Contact had the best music. And the music from the JJ Trek movies was pretty good also, with "Enterprising Young Men" and the end credits adaptation of the original series' theme being favorites for me.
And if we're considering video game scores, go give the soundtrack from Starfleet Command II/Starfleet Command: Orion Pirates a listen sometime. You're welcome.

As much as I want to give the nod to TNG's opening theme given I grew up with it, I'm going with The Wrath of Khan's music. The opening credits overture is great.

If I were going to put something that was my favorite but definitely not the best theme in the mythos, I would put the opening Stellar scene from Star Trek the motion picture where it's just the stars going away from you while it's symphonically whimsically escorting you to space. That sounds flowery the way I put it, but that's how it makes me feel
I'm firmly in the Klingon camp as well.
Second would be the beastie boys, because I like them even if I thought it was not setting appropriate imop but to be fair that, to me, was the best part of that particular movie.
The rewards of tolerance are treachery and betrayal.
Remember kids, Games Workshop needs you more than you need them.
But, yeah, the TNG theme is first for me, followed by the Klingon theme and then James Horner’s theme(s) for 2 and 3.
Oh, it was. Time signature changes, key changes, but the motif is always there when the Klingons are around. I only started to really notice it by Enterprise, during the Augments arc.
Then I went back and rewatched TNG and DS9, and it was there. Any Klingon heavy story used the motif.

The klingon suite is my #1
The soundtracks (deluxe editions) for ST II the wrath of khan and STVI the undiscovered country are my favorite official releases
As an added bonus he did a collection of all into intro themes.

But seriously, I'd have to go with the score for The Motion Picture. The film was released before it was ready, much like the Enterprise and her crew in the film, but the score was bloody incredible and help made V'Ger the most chilling force any of the crews have ever faced, as well as the most awesome. If I had to pick just one moment from the whole film, it would be from the following track at 3:15...
