Recently, I've started watching Death Note and Fullmetal Alchemist: Brotherhood. I have to say I'm sad that I have spent so long before watching Brotherhood, because it is truly an amazing anime. It's so much better than the first Fullmetal Alchemist series they did.

Well Azumanga Daioh is one, and while it does have few A) moments, School Rumble is also somewhat good choice.
Yea. If you let some of the humour ease out the bit more 'depressing' bits, theirs a lot out their. If you’re looking to get into anime, try starting with bleach. Full of funnies and well, hasn’t finished yet, but they did beat the big bad.

Third. I love what they did. They made Vegeta sound more awesome. And Making fun of Krillin is awesome.. By far this is my most favorite episode of the Abridged Series.
All the voice actors are awesome and do it very well

Project A-Ko is fun, ludicrous, and quite the opposite of depressing.
Wow, if that one doesn't bring me back...
Tengen Toppa Gurren Lagann pretty much defines epic feel-good ending for me, although the very end is a bit bittersweet. It has some darker moments throughout, too, so might not fulfill your requirements to a t, but it hasn't yet failed to get me excited to be alive when I finish the series.

Spice and wolf
The only reason Spice and Wolf doesn't have a sad ending is because it isn't over yet. It's also got more depressing Fridge Logic (and not-so-Fridge logic) than you can shake TVTropes at. That being said, it's pretty awesome.
I'd personally recommend K-On! and K-On!! (read: season 2). So sweet it'll give you diabetes.
