Alright when doing The Joker from Batman (any media) should his smile be from some disfiguration or should it just be make up? Bonus: Skin is colored white or just uses white make up?

As a DCAU fan, I always prefer that the joker's skin is actually bleached white and his smile is from disfiguration, so his lips are stark red naturally, not from lipstick.

It helps tie into how the Joker persona is etched into his very being and not something he puts on/off like Batman does with his mask.

The character he’s based on from The Man Who Laughs is disfigured with a permanent smile. I say If it ain’t broke don’t fix it.

Skin & hair should be dyed white/green.
Because falling into the vat of Ace Chemicals is his origin.

The lips/smile? I'd generally argue for the Ace chemicals But if (probably self inflicted) disfiguration + makeup works for the particular story.
Afterall, he is crazy. So if he comes home dyed all white & green, looks in the mirror, and thinks he now looks like the Joker from a deck of playing cards....

hair and face color permanent for sure.

I'm torn on the smile. I think I prefer it not to be a disfiguration and just naturally, unnaturally wide. The ability for the smile to grow out from a fairly normal face is super unsettling and core to a lot of my favorite Joker moments.

The tatted up Juggalo for a Joker isn't my favorite take. However, gotta be honest, it kinda works along side of how they did Harlie Quinn. His appearance in the Snyder cut of Justice League was pretty good.

Re: Leto, I thought his portrayal was too hammy and the tats were overdone. Pull those both back by half and I think it might have been interesting though.

The slicked-back hair did evoke the Joker's look in the comics at the time. And there's nothing inappropriate about a move to more of a straight-up criminal and away from a nihilistic terrorist as in the Nolan films. As I've discussed many times before, the Joker has been very different things in the comics at different times...much moreso than other characters to the point where some writers have tried to explain it (Grant Morrison chalked it up to a kind of adaptive 'supersanity' where the Joker constantly reinvents himself, while Geoff Johns suggested that there have been three different Jokers).

So I think trying different things with the Joker is in a way the most source material-based thing one can do. To me, saying the Joker "should" be this or that way just kinda shows one isn't very familiar with the totality of the character.

Regarding the OP's question, I think the answer is that it should be whatever works for the character at that given time. Call it a cop out if you want, but to me the Joker is just an extremely protean character whose details are best left malleable to match.

Thing is, we had a near perfect live action trifecta.

Cesar Romero was the straight up goofy goober Joker, as suited the “kids comic” feel of the Batman TV Series.

At completely the other end of the spectrum we have Heath Ledger’s portrayal. A twisted psychopathic anarchist.

And as a comfortable stepping stone between the two, Jack Nicholson’s portrayal which kept some of Romero’s comedic portrayal, but essentially redefined Joker as a scary villain for many people, especially those like myself who’ve never really read the comics.

Leto’s portrayal felt like a love letter to himself. A celebration of how wonderful he thinks he is. Overly edgy. Not at all menacing. Just a tosspot. The bit that really set me off was the stupid grin on his stupid hand and holding it up to his stupid bloody face then doing that stupid fake laugh. Even the guy he was about to kill didn’t look intimidated so much as seemed to be thinking “what a knob”.

We also have Hamil’s Joker. However, his time in the smile has been varied, depending on which media he’s provided the voice for.

Don't think of it as "should" think of it as you being given the task of doing a Joker story and having creative freedom to choose the look you want for that story. I don't think there is any confusion to the fact there have been different takes on the character this is about your take.
